The Javelina Jundred is run in loops on the Pemberton Trail, inside McDowell Mountain Regional Park. Every lap follows the same circuit, and the poster traces one of them. Start and finish share the same point, the Four Peaks Staging Area — also known as Javelina Jeadquarters. The gun goes off at 6 a.m.

From the start, the trail climbs steadily along the Pemberton Trail to Jackass Junction, the loop's high point. The route then drops back west across the park to Wagner Trailhead, before closing the loop to the south. The ground alternates between the Pemberton, Shallmo and Cinch Trails, through the Sonoran Desert, inside McDowell Mountain Regional Park (Maricopa County Parks).

The 100 Miles repeats this loop four more times, after a longer opening lap that takes the Escondido Trail before rejoining the regular route. Runners get 30 hours to complete the whole distance.
